OBJETIVO: Investigar la actividad de la catalasa en el metabolismo de tres tejidos diferentes: hígado, riñón y pulmón.
MÉTODO:
Se monta el dispositivo (ver ilustración), mediante el cual se puede medir con exactitud la cantidad de O 2 desprendido en la reacción.
Se añade 1 g de cada una de las sustancias al mortero, se le añade un poco de arena y se tritura. Se añaden 10 cm 3 de agua destilada y se mezcla bien.
Se vierte 2 cm 3 de la mezcla anterior en un matraz.
En una especie de “tapón de rotulador” pero graduado, se añaden 2 cm 3 de H 2 O 2 y se sujeta mediante un hilo. Se introduce dentro del matraz, pero sin que se caiga sobre la mezcla que ya habíamos introducido.
Cerramos bien el matraz, dejamos caer el H 2 O 2 en la mezcla y esperamos 30 s. Anotamos la cantidad de O 2 desprendido.
Repetimos este proceso de 2 a 3 veces con cada tejido.
